Genetica e Miglioramento Genetico Per l'Allevatore

- Lo Studente deve possedere le nozioni fondamentali che lo mettano in condizione di comprendere:

  • i concetti alla base della Genetica classica, delle modalità di trasmissione dei caratteri
    ereditari, della struttura fine del gene e della Genetica molecolare per le specie animali in
    produzione zootecnica e le specie animali da compagnia.
  • i concetti alla base della Citogenetica: assetti cromosomici normali ed anomali delle specie
    animali in produzione zootecnica e delle specie da compagnia.
  • le moderne implicazioni della genetica molecolare nelle metodologie di
    valutazione genetica dei riproduttori per il miglioramento genetico  per la corretta applicazione degli schemi di selezione genetica negli animali da compagnia;
  • le moderne implicazioni della genetica molecolare in campo clinico nello studio delle patologie
    a base genetica mono e multi fattoriale del Cane del Gatto con la finalità di comprendere i meccanismi di eradicazione delle patologie mono fattoriali attraverso la precoce individuazione dei riproduttori affetti, sani e portatori sani;
  • le Applicazioni della Genetica Molecolare e le procedure da seguire nel rilascio dei Pedigree
    da parte dell’Ente Nazionale per la Cinofilia Italiana E.N.C.I.: Deposito e conservazione del
    Campione Biologico presso Laboratori accreditati, Profilo Genomico mediante marcatori
    Genomici STR, Test parentale (Esecuzione ed Interpretazione).

The student must possess the fundamentals that make him able to understand:
the basic concepts of classical genetics, , fine gene structure and molecular genetics for animal species in livestock production and pet species.
the basic concepts of Cytogenetics: normal and abnormal chromosomal patterns of the species
animals in zootechnical production and of the species of companionship.
The modern implications of molecular genetics in the methodologies of genetic evaluation of breeding animals for the genetic improvement and for the correct application of genetic selection schemes in pet species;
The modern implications of molecular genetics in the clinical field in the study of pathologies
Cat and Dog Factor Mono and Multi Factorial Genetic Base with the aim of understanding the mechanisms of eradication of pathologies
mono fatty acids through the early detection of affected, healthy and healthy carrier reproducers;

the Applications of Molecular Genetics and the procedures to be followed in the release of Pedigree
by the Italian Kennel Club E.N.C.I .: Deposit and Conservation of the

L'accertamento delle conoscenze avverrà tramite prova scritta su piattaforma informatizzata. La prova verterà sugli argomenti trattati nel corso delle lezioni frontali e delle esercitazioni pratiche svolte in laboratorio Informatico su Piattaforma Moodle dell' Università di Pisa. (Secondo le Modalità stabilite dall' Università di Pisa per il distanziamento e la protezione degli Studenti dal Covid 19)

Knowledge assessment will be carried out through a written test on a computerized platform. The test will focus on the topics discussed during the frontal lessons and practical exercises carried out in the Computer Laboratory at Platform Moodle of the University of Pisa in presence. (According to the procedures established by the University of Pisa for the distancing and protection of students from Covid 19)

Genetica e Miglioramento Genetico Per l'Allevatore

  • Lo studente sarà in grado di applicare i modelli di trasmissione ereditaria per caratteri semplici e complessi e di individuare i casi ererditari dovuti alle estensioni delle leggi di Mendel.
  • Lo studente sarà in grado di prelevare e conservare i campioni biologici sia ai fini del depistaggio delle malattie genetiche che delle analisi richieste dell’Ente Nazionale per la Cinofilia Italiana E.N.C.I. per le le procedure da seguire nel rilascio dei Pedigree e sarà in grado di interpretare il referto di un test parentale.
  • Lo studente sarà in grado di impostare un percorso di tracciabilità individuale e Razziale lungo tutta la filiera Produttiva Carne e Latte e sarà in grado di applicare le metodiche Probabilistiche e Deterministiche di tracciabilità nelle rispettive filiere produttive Carne e Latte.
  • Lo studente sarà in grado di applicare i moderni schemi di selezione genetica, saprà utilizzare i test di genetica molecolare nel depistaggio delle patologie del Cane del Gatto e delle principali specie in produzione zootecnica, ai fini dell’eradicazione delle patologie mono fattoriali attraverso l'individuazione precoce dei riproduttori affetti, sani e portatori sani;

 

Skills

 

Genetics, Animal Breeding and selection For the Breeder

The student will be able to apply inherited transmission models for simple and complex characters and to identify hereditary cases due to extensions of Mendel's laws.
The student will be able to pick up and store the biological samples both for the purpose of screening for genetic diseases and for the required analyzes of the Italian Kennel Club E.N.C.I. for the procedures to be followed in the release of Pedigree and will be able to interpret the report of a parental test.
The student will be able to suggest to the breeder the DNA tests for prevention in dogs and cats. DNA tests for disease-causing genes allow breeders to avoid breeding dogs that could result in puppies being affected by the disease and to improve the health of their breeding stock.
The student will be able to apply the modern genetic selection schemes and will use molecular genetic testing in the screening of Pathologies for the purpose of eradicating monofactorial pathologies.

Verifica dei test parentali effettuata mediante simulazioni computerizzate. Piattaforma Moodle dell’Università di Pisa.

Riconoscimento delle Razze canine ed attribuzione ai 10 Gruppi FCI mediante attività interattiva ed auto valutazione dell'apprendimento  eseguita presso il "Polo Informatico 5 Piagge" mediante piattaforma informatizzata sistema MOODLE dell'Università di Pisa. Possibilità da parte dello Studente sia di esercitarsi in questa attività che di auto valutare la sua preparazione anche da remoto con qualsiasi device (PC, Tablet, Smartphone)

Practical activity at the "Polo Informatico 5" laboratory using the MOODLE system computerized platform of the University of Pisa. Constitution of a genomic profile by Microsatellite and SNP Genomic Markers as the ISAG International Society for Animal Genetics Protocol and Attribution of a genomic identity to an individual or biological sample for the purpose of recognition in the event of an expert report or certification of seminal material for the purpose of artificial insemination. Verification of parental tests carried out using computer simulations. 6h

Recognition of dog breeds and attribution to the 10 FCI Groups through interactive activities and self-assessment of learning performed at the "Polo Informatico 5 Piagge" through the computerized platform of the University of Pisa's MOODLE system.2h

Genetica per l'Allevatore

(4h) Presentazione del programma del Corso di Genetica. Richiami sulla struttura, composizione chimica, e funzione degli acidi nucleici. L’organizzazione del DNA nei cromosomi. Dimensione dei Genomi delle Specie animali in produzione zootecnica e da compagnia. Contenuto del DNA Ripetuto, DNA a Sequenza Unica ed a Sequenza Ripetuta. Replicazione e Ricombinazione del DNA. Il Gene: struttura chimica, meccanismi d'azione, criteri di studio. La Trascrizione. Il codice genetico e la traduzione. La regolazione dell'espressione genica. Mutazioni geniche, mutazioni soppressore, tasso di mutazione, frequenza di mutazione. Come le mutazioni in un gene possono influenzare la sua espressione. Genetica molecolare Nozioni di base.

La Genomica applicata alla tutela delle razze canine. Caso di Studio: l’importazione illegale di cuccioli. La Genomica forense applicata alla specie canina. (4h) Le basi genetiche della taglia e della conformazione corporea del cane. L'esempio dello studio effettuato nella razza Canina Portuguese Water Dog. Il Caso particolare del gene MSTN nella razza Canina Whippet. Associazioni tra dimensioni corporee e mutazioni presenti nei geni IRS4, ACSL4 e IGSF1 nelle diverse razze canine. 

2h) Le basi genetiche della longevità del cane e la loro correlazione con la Taglia Corporea. La correlazione tra lunghezza dei telomeri e la longevità. QTL CFA 7; QTL CFA 8; QTL CFA 10; QTL CFA 15; QTL CFA 34; QTL CFA 23; QTL CFA 29; QTL CFA 9.  

2h) Lo studio della base genetica delle malattie ereditarie del Cane. Management delle Malattie Genetiche. Anomalie cromosomiche. Patologie monogeniche ed approcci di studio. Patologie multifattoriali, multigeniche ed approcci di studio.

2h) Le Basi Genetiche dell'Olfatto del Cane. Il Polimorfismo dei geni dei ricettori olfattivi del cane. Il Ruolo dei Marcatori Genomici SNP e del loro Polimorfismo nella capacità discriminatoria delle diverse molecole odorifere. Il polimorfismo dei marcatori genomici SNP correlato con l'attitudine innata nel riconoscere particolari molecole olfattive. 

(2h)  Il ruolo dei  geni IRS4, ACSL4 e IGSF1 nelle dimensioni corporee delle razze con peso maggiore ai 40Kg. I geni alla base della conformazione dell'orecchio e della lunghezza ed arricciamento della coda. I geni che influenzano le caratteristiche del Mantello. Il gene FOXI3 nella razza Chinese Crested. Mutazioni sul CFA18 e mantello della razza Rhodesian Ridgeback. Il gene RSPO2 e la caratteristica Furnishing nelle razze Schnauzer e Scottish Terrier. Il ruolo dei geni FGF5, RSPO2, KRT71 nelle razze nude. Il gene SGK3 e la non letalità nella razza nuda American Hairless Terrier analisi del pedigree

Stato dell'arte della mappa genomica delle specie da compagnia. RICERCA DI QTL MEDIANTE L’IMPIEGO DI MARCATORI GENOMICI.   

(4h) IDENTIFICAZIONE DEI GENI RESPONSABILI DELLE PRINCIPALI MALATTIE EREDITARIE. Approccio dei MARCATORI MOLECOLARI per la ricerca delle basi genetiche delle patologie multifattoriali, Epilessia, Displasia dell'anca etc.  Approccio del Gene Candidato il caso della PRA. Analisi Genome Wide Association Scan. Confronto analisi di Linkage analisi GWAS. l'esempio della PRA Atrofia Progressiva della Retina del cane. Malattie Tumorali. Le basi genetiche dei tumori correlati alla selezione attuata per il colore del mantello e della taglia corporea. il ruolo dei Test Genetici nell'allevamento del cane. Messa a punto di un test genetico ed utilizzo dei test genetici nella prevenzione della Comparsa di una Malattia a base genetica e nell'eradicazione della malattia in un allevamento.

Aberrazioni cromosomiche. Alterazioni del numero: polisomie e poliploidie. Non disgiunzione meiotica e mitotica. Anomalie di struttura. Delezioni. Duplicazioni. Il Concetto di Specie ed il concetto di Razza. Richiami su come sono state create le moderne razze canine. Significato di Pool Genico. Il Pool Genico e suo utilizzo nella fondazione di una Razza. Genetica di Popolazione. Come cambia la Variabilità Genetica nel tempo e nello spazio. Determinazione della vulnerabilità genetica di una razza canina utilizzando i criteri sviluppati dalla .  2h

Comprendere la composizione genetica di una popolazione e le forze che determinano e modificano tale composizione. Genetica di Popolazione. Concetto di Frequenza Allelica e Frequenza genotipica. La Legge di Hardy Weimberg. Test dell’equilibrio di Hardy-Weinberg. 2h

Come si spiega la Variabilità genetica di una popolazione in termini di Meccanismi Genetici: selezione sessuale, accoppiamenti assortativi positivi e negativi nelle popolazioni naturali  e confronto con gli accoppiamenti selettivi nella specie canina il concetto di Inbreeding, esempi sul cane. La Deriva Genetica, La Mutazione, Mutazioni Sinonime e non Sinonime, La Migrazione. 2h

L’ effetto “Collo di Bottiglia”, l’effetto Fondatore” Esempi condotti sulle razze canine. L’effetto sulla variabilità genetica delle popolazioni canine dei riproduttori “più Popolari e Vincitori di Gare di Bellezza o Bravura”. Concetti di Dimensione Effettiva di un Popolazione e di Numero effettivo. La Statistica di F di Wright e l’analisi della variabilità genetica nelle popolazioni 2h

I diversi obiettivi della Selezione adottati nelle razze Canine. Gli Schemi selettivi adottati nella Specie Canina ed i loro effetti sulla variabilità Genetica e fenotipica. Perchè è fondamentale individuare l’azione della selezione a livello genomico. 2h

I Libri genealogici. I pedigree. Concetti di Parentela e Consanguineità (Inbreeding) perché è importante conoscerli a scopi selettivi e per la salute genetica delle razze canine. Le misure di Base della Parentela, il Coefficiente di Kinship, Coefficiente di Wright. Calcolo Della Parentela Additiva e della Consanguineità: Il Metodo del “tracciare le Vie”, Il Metodo Tabulare, La Matrice di Parentela Additiva. Esempi sulle razze canine. Marcatori Microsatelliti (STR) e SNP e Parentela Genomica. 2h

I Caratteri Quantitativi nella specie canina. Classi fenotipiche di caratteri quantitativi e distribuzione dei fenotipi. Applicazione dei Concetti di: Media, Varianza e Deviazione Standard.
